In vitro study | Danshensu can reduce lipid peroxidation on mitochondrial membrane by scavenging free radicals, reducing sulfhydryl oxidation, inhibiting mitochondrial membrane permeability and transport. Danshensu can significantly improve the cell viability of H9c2 cardiomyocytes and reduce the release of LDH. In cells, increased phosphorylation of Akt and ERK1/2. The protective effect of danshensu can be PI3K inhibitor wortmannin or ERK inhibitor u0126. Danshensu can exert its cardioprotective effect in ischemia-reperfusion model, and the possible mechanism is to inhibit cardiomyocyte apoptosis. Danshensu increases the expression of Bcl-2 and reduces the expression of Bax and active caspase-3 by activating Akt and ERK signaling pathways. Danshensu can play an active role in many biological processes, such as improving microcirculation, inhibiting the production of reactive oxygen species, inhibiting platelet adhesion and aggregation, protects endothelial cells from damage caused by inflammation. |
In vivo study | Pretreatment of ISO-treated rats with danshensu significantly reduced the ST segment (P<0.001). Its pretreatment can also significantly reduce the level of cTnI in serum. Therefore, Danshensu can have a protective effect on myocardial infarction in rats induced by ISO. In the rat model of myocardial ischemia-reperfusion injury, Danshensu significantly reduced the area of myocardial infarction, the production of CK-MB and the level of cTnI in serum. |
